880 3 6| 520-00/(Sa| Over-expression of the proto-oncogene B-cell lymphoma {Bcl-2) has been found in a wide range of cancers including breast. Bcl-2 encodes an integral outer mitochondrial membrane protein that blocks apoptosis. In addition, the induction of Bcl-2 expression has been associated with the retardation of mammalian cell proliferation due to the lengthening of the G1 phase. We are interested in observing the gene expression changes that occur in cells over-expressing Bcl-2. We hypothesized that the over- expression of Bcl-2 gene will affect the regulation of the genes that control the cell cycle. We should observe the down-regulation of genes that allow the progression of the cell cycle and the up-regulation of those that block the progression. The gene expression profiles of two cell lines, MCF10A/Bcl-2 and control MCF10A/Neo, were compared using cDNA microarrays. A total of 363 genes were identified as differentially expressed by at least two-fold; 307 were up-regulated and 56 down-regulated. Using microarray data and pathway analysis tools, target genes were selected for further analysis. Parametric gene set enrichment (PAGE) identified 22 significantly changed gene sets, containing genes that function in regulating cell proliferation, apoptosis, blocking cell cycle progression, inducing DNA repair, and insulin signaling. The highest ranked network identified by Igenuity Pathway Analysis (IPA) contained 24 significant genes. This network included genes whose products function in controlling the cell cycle, cell proliferation, metastasis, and angiogenesis. Using qRT-PCR we confirmed the up-regulation of two cell cycle genes, Ccnd1 and Cdc42 and the down-regulation of Cdc25A. We also confirmed the differential expression of six other genes involved in proliferation, cancer progression, and invasiveness. The down-regulated genes were Akr1C3, Tgfβ1, and Cyr61, while the up-regulated genes were Mmpl, Fnl and Tgfβ3. We concluded the increased levels of Cdc42 and Ccnd1 could be functioning to slow the cell cycle but the exact mechanism is still unclear. However, the down-regulation of Cdc25A alone could be responsible for the slow cell cycle progression observed in the MCF10A/Bcl-2 cells.;
