Summary Previous studies have identified a number of proteins in extracts of the x-organ/sinus gland complex in a variety of Crustacea and other arthropods. In several species, substances similar to the vertebrate vasopressins have been reported. The goal of this study is : 1) to examine the distribution of proteins in extracts of blue crab sinus glands, and 2) to investigate the presence of vasopressin-like peptides in the sinus gland of the blue crab, Callinectes sapidus. Crabs were collected from the Pamlico and Albemarle Sounds of North Carolina. Eyestalks of the crabs were removed and stored at -70°C. Proteins were extracted by boiling in 0.1N HCl and filtering through Sep-Pak C-18 columns. Samples were then subjected to HPLC and SDS-PAGE. Other eyestalks were dissected to remove the sinus glands which were then extracted as above. Extracts were subjected to SDS-PAGE and HPLC. Results of these experiments indicate the presence of 11 eyestalk proteins. Sinus gland extracts contained a factor that co-migrated on SDS-PAGE and co-eluted from HPLC with lysine- and arginine-vasopressin. Utilizing the criteria established by Mattson & Spaziani (1985) and others, it is possible that this factor is a blue crab molt-inhibiting hormone (MIH).
