Summary The present study uses lectin histochemistry to examine the extent to which prolactin (PRL)-induced integumental changes in red-spotted newts are due to alterations in the expression and distribution of glycoconjugates within the epidermis and epidermal glands. A panel of eight horseradish peroxidase labelled lectins (LTA, UEA-I, DBA, SBA, GSAI-B4, WGA, PNA, CON A), which bind specifically to terminal carbohydrate moieties of glycoproteins and glycolipids was chosen. The epidermis of PRL-injected newts showed a marked decrease in lectin binding for LTA, DBA, SBA, and PNA as compared to control salineinjected newts. The binding of WGA to sialic acid increased with PRL injection, while GSAI-B4 and CON A showed little to no change in binding. Temporal studies revealed that differential lectin binding occurred by day 6 in PRL-injected newts a well as the epidermis of freshly collected newts undergoing conditioning. Seasonal examination revealed similarities in the expression and distribution of epidermal glycoconjugates between freshly collected winter animals and laboratory conditioned animals that had received PRL. Together, these findings demonstrate that prolactin may bring about integumental changes in the red-spotted newt by affecting the expression and distribution of both secretory and structural glycoconjugates.
