Summary Studies were undertaken to determine the effects of light upon the movements of sodium ions within isolated rod-outer-segments of cattle and excised whole frog retinae. All experimentation was under darkroom conditions at 0-4°C. Cattle rod-outer-segments were Isolated and concentrated in solutions of 0.32M sucrose. A parallel set of experiments incubated the rod-outer-segments in Ringer solution containing Na22 Aliquots of the suspension were taken and half were illuminated, others remained non- illuminated. Following illumination and an interval of incubation, the suspensions were centrifuged. The supernatant was separated from the rod-outer-segments and analyzed for sodium present by flame photometry and tracer technique. Single whole frog retina was excised and equilibrated in a flowing Ringer solution containing Na22 . The retina was then transferred to a specialized chamber through which normal Ringer solution passed and collected at specific time intervals as aliquots. Illumination of the retina occurred for some duration during the elution. The resultant data were analyzed by visual and statistical examination. Results of whole retinae experiments indicated that sodium efflux occurred at some time after illumination. The rod-outer-segment experiments indicated the efflux occurred as early as two minutes and ceased prior to fifteen minutes after illumination.
