Summary This study was designed to address the morphological effects of different storage media on the corneal epithelium. Optisol GS is the intermediate term maintenance medium used by the majority of eye banks in the United States today to preserve both endothelial and epithelial integrity of the cornea. Epithelial morphology was evaluated using a scanning electron microscope after storage in either Optisol GS or Optisol GS enhanced with Vitamin E and Dexamethasone for 3, 5, or 10 days. With permission obtained from the North Carolina Eye and Human Tissue Bank, corneas were retrieved from adult human cadavers and stored for varying lengths of time in each medium. The recovery and storage of the corneas followed the standards set forth by the Eye Bank Association of America. Representative photographs were taken of the corneal epithelium and used to determine the amount of sloughing for each cornea. A percentage of sloughing vs. normal cells was obtained and statistically analyzed. The results of this study determined that the corneas stored in Optisol GS supplemented with Vitamin E and Dexamethasone had significantly less epithelial sloughing than those stored in Optisol GS.
