880 3 6| 520-00/(Sa| In Lewis rats, MBP-specific T-helper (Th) cells mediate the demyelination of central nervous system axons in experimental autoimmune encephalomyelitis (EAE). EAE serves as a relevant animal model of the human demyelinating disease multiple sclerosis. Adoptively transferred encephalitogenic Th cells have been found to induce resistance to EAE in Lewis rats. The mechanism of this resistance is not known, though several reports have attempted to link resistance to Vβ gene expression of encephalitogenic Th cells. The purpose of this study is to examine the Vβ expression of encephalitogenic Th cells and the relevance of Vβ expression to the induction of resistance. Several polyclonal lines derived from Lewis rats sensitized with R72-86 peptide were tested for Vβ expression. Furthermore, a number of monoclonal lines specific to various determinants of MBP were tested for the ability to induce resistance to EAE. The results of this study reveal heterogeneity among encephalitogenic T cells specific to MBP68-86 with regard to V β expression and CDR 3 motif. Encephalitogenic Th cells expressing Vβ4 and Vβ10 established resistance to EAE induced by the injection of GPMBP in CFA. Nonencephalitogenic Th cells bearing Vβ3 and Vβ8.6 did not, however, effectively protect Lewis rats from EAE. The results of this study demonstrate that the induction resistance to EAE does not require incorporation of particular Vβ genes by encephalitogenic TCR p chains.;
