| Summary | The purpose of this study is to determine the effects of chronic estrogen replacement on heat shock protein 70 (HSP70) expression in the heart. To test this hypothesis. New Zealand White rabbits underwent bilateral ovariectomy after-which, they were randomly divided into several groups: controls, ovariectomized (OVX) rabbits without hormone replacement with 17-P estradiol, OVX with low-dose hormone replacement (50pg/kg bodyweight/day) and OVX rabbits receiving high-dose hormone replacement (90pg/kg bodyweight/day). Hormone replacement occurred over the course of 60 days. The mean plasma estradiol levels were 130 picograms/ml (pg/ml) in controls, 156 pg/ml in OVX rabbits without hormone replacement, 249 pg/ml in OVX rabbits receiving low-dose hormone replacement and 520 pg/ml in OVX rabbits receiving high dose estrogen replacement. Isolated heart studies were performed to determine the effects of estrogen on ischemia and ischemic preconditioning. Western blot analysis revealed a significant change in HSP 70 expression in response to increased plasma estradiol levels obtained via hormone replacement therapy, with 17-P estradiol. In addition, increases in plasma estradiol levels correlated with significant reductions in infarct size. The results from this study demonstrate that the presence of estrogen leads to an increase in HSP 70 expression. These findings suggest that the increase in HSP 70 may potentially represent another mechanism whereby estrogen replacement therapy leads to enhanced cardioprotection in post-menopausal women. |
