880 3 6| 520-00/(Sa| The purpose of this study was to test the hypothesis that decreased fatty acid oxidation by the muscle of obese subjects leads to an increase in ketone body oxidation in order to supply the appropriate amount of energy needed by the muscle. Ketone body metabolism was studied in: 1) obese and non-obese women; 2) ob/ob obese mice and their lean littermates; and, 3) leptin treated ob/ob obese mice (post-obese) and their lean littermates. Plasma ketone body concentrations were measured and the capacity of skeletal muscle homogenates to oxidize β-hydroxybutyrate were determined in the human and animal models. In addition, the rates of CO2 and acid soluble metabolite production from fatty acid oxidation by liver homogenates were determined from lean, obese and post-obese mice. Mitochondrial oxidative capacity was determined by measuring the activities of citrate synthase and β-hydroxyacyl-CoA dehydrogenase. The results showed that the plasma ketone body concentration and the β-hydroxybutyrate oxidation by muscle were decreased in the obese human subjects as compared to the non-obese controls (P<0.05). The concentrations of plasma β-hydroxybutyrate in the lean, obese, and post-obese mice were not different. However, the rates of acid soluble metabolite production from hepatic fatty acid oxidation and the rates of β-hydroxybutyrate oxidation by muscle of the obese mice were higher (P<0.05) than that of the lean controls. Conclusions: 1. The ob/ob obese mouse is not a good model for comparison with humans when examining ketone body metabolism; 2. Ketone body oxidation in skeletal muscle of obese humans does not compensate for the decrease in fatty acid oxidation in this tissue. Muscle of obese humans appears to have a decreased capacity to oxidize substrates. One explanation for this may be the decreased mitochondrial activity in the muscle of the obese subjects as evident from decreased citrate synthase activity.;
